Guanidine HCl or isothiocyanate

Decide whether you need to unfold protein or release nucleic acid.

Guanidine hydrochloride (CAS 50-01-1) is a strong protein denaturant. About 6 mol/L is a common working concentration for unfolding and inclusion-body solubilization. Guanidine isothiocyanate (CAS 593-84-0) is also a strong chaotrope. The guanidinium cation denatures proteins, including nucleases. The thiocyanate anion makes it a stronger chaotrope than the hydrochloride, which is why nucleic-acid protocols more often specify it. Catch-Chem quotes both as solids; grade follows the batch COA.

Does thiocyanate by itself inhibit nucleases?

The guanidinium cation denatures proteins, including nucleases. Thiocyanate is what makes GITC the stronger chaotrope.

Can the two salts replace each other?

No. The CAS numbers and uses differ.
