G418 selects eukaryotic cells that carry neo/kan. Activity is an active concentration, so a new lot needs a kill curve. Do not treat the bottle weight as the working concentration, and do not substitute this salt for bacterial kanamycin.

What it is

G418 (Geneticin) is an aminoglycoside related to gentamicin. It binds the 80S ribosome and blocks elongation, killing eukaryotic cells. The neo gene (aminoglycoside 3'-phosphotransferase, APH(3')-II) inactivates it by phosphorylation, so only transfected cells survive.

Where it is used

  • Stable mammalian cell lines: typically 200–800 µg/mL active G418 for selection, often about 400 µg/mL, over roughly 1–3 weeks.
  • Maintenance of established lines at a reduced dose, commonly about half the selection level.
  • Yeast selection with kanMX cassettes, often around 200 µg/mL.
  • Plant and other eukaryotic systems carrying nptII. Set-up is covered in how selection is set up.

Handling tips

  • Calculate the dose on active G418 (µg active per mg on the COA), not on powder weight.
  • Run a kill curve for each cell line and each new lot.
  • Prepare a stock around 50 mg/mL active in water or buffer, filter-sterilize, and store at 2–8 °C or −20 °C.

What to specify when ordering

  • Potency (µg/mg) and cell-culture grade.
  • Powder or sterile solution with concentration.
  • Pack size.

Before you inquire

Do you assay the potency?

No. The supplier COA is forwarded.

Is a 20 mg/ml solution available?

Powder and sterile solution can both be inquired. For a solution, give the concentration.

Can G418 select bacteria?

It is not the usual choice. Bacterial neo/kan plasmids are selected with kanamycin, which is on the antibiotics page.
