Not the same as
A 1:4000 mixed digestive enzyme

This is the USP 1:250 trypsin used to passage cells and, in proteomics, to digest proteins. It is a different product from a 1:4000 mixed digestive enzyme. For cell culture, consider whether EDTA is used and how the enzyme is inactivated.

What it is

Trypsin is a pancreatic serine protease that cleaves peptide bonds on the carboxyl side of lysine and arginine, except before proline. The USP 1:250 designation describes a porcine pancreatic preparation by its digestive strength; it also carries other pancreatic proteases, which helps in dissociating cells.

Where it is used

  • Detaching adherent cells for passaging, typically 0.25% or 0.05% trypsin, often with EDTA (about 0.02%, 0.5 mM), for a few minutes at 37 °C.
  • Dissociating primary tissue into single cells, sometimes with collagenase.
  • Bulk protein digestion. LC-MS peptide mapping usually calls for TPCK-treated or sequencing-grade trypsin, which is a different specification.

Handling tips

  • Rinse cells with calcium- and magnesium-free PBS or DPBS first; divalent ions and serum inhibit detachment.
  • Stop digestion with serum-containing medium or soybean trypsin inhibitor; over-exposure damages surface proteins.
  • Filter-sterilize solutions (do not autoclave), aliquot and store at −20 °C; avoid repeated thawing.

What to specify when ordering

  • Grade: USP 1:250 or a higher-purity grade, and source species.
  • Powder or sterile solution, with or without EDTA and phenol red.
  • Pack size. Context: cell culture.

Before you inquire

Is this 1:4000?

No. The listed item is USP 1:250.

0.25% or 0.05% trypsin?

0.25% works faster for strongly adherent lines. 0.05% is gentler and common for sensitive or routinely passaged cells. Titrate time against detachment.

Is this suitable for mass-spectrometry digestion?

Crude 1:250 trypsin contains chymotrypsin and other proteases. For LC-MS, ask for TPCK-treated or sequencing-grade trypsin.
